Analysis of the extraction of bioactive peptides obtained from collagen in marine species (fish and crustaceans): Systematic review

Abstract

The valorization of marine by-products and the growing demand for functional ingredients have positioned bioactive peptides derived from collagen as compounds of high interest due to their therapeutic properties and applications in the food industry. This article aims to systematically analyze the extraction methods, structural characterization, and functional evaluation of bioactive peptides obtained from the collagen of marine species, highlighting their technological feasibility and potential within circular bioeconomy frameworks. A total of 72 studies published between 2020 and 2025 were reviewed following PRISMA 2020 guidelines, focusing on antioxidant, antihypertensive, antimicrobial, and osteoprotective activities. Collagen extracted from structures such as dermis, scales, and exoskeletons has emerged as a sustainable source for generating functional peptides. The most studied species include cod (Gadus morhua), tuna (Thunnus albacares), flounder (Paralichthys olivaceus), shrimp (Litopenaeus vannamei), and crab (Portunus trituberculatus). Emerging technologies such as ultrasound-assisted extraction (UAE), microwave-assisted extraction (MAE), and high-pressure processing (HPP) enable the production of low molecular weight peptides (<3 kDa), associated with enhanced bioactivity. In vitro studies show angiotensin-converting enzyme (ACE) inhibition (IC₅₀: 0.05–0.25 mg·mL⁻¹), high antioxidant capacity (>70% DPPH), and metal chelation, supporting their potential as functional food ingredients.
